Acetaminophen (AST), if taken in excess, causes nausea, jaundice, hepatotoxicity and acute hepatic failure. To evaluate the hepatoprotective effects of Astragalus ecbatanus hydroalcoholic extract against repeated acetaminophen-induced liver injury in male Wistar rats. Repeated acetaminophen-induced liver injury was established by oral administration of acetaminophen (300 mg/kg, twice a week for 8 weeks). Following this procedure biweekly for a period of 8 weeks, the rats received injections of 25 and 50 mg/kg of the extract. Subsequently, we quantified the levels of LPO, SOD, CAT, GPX, AST, ALT, ALP, bilirubin, and total protein in sera and LPO in liver tissues, which were considered to be biochemical indices of OSI and LDI. The result of administering AST indicated a significant rise in the serum concentration of LDI (ALT, AST, ALP, bilirubin, and total proteins), MDA levels, and a significant decrease in the activity of SOD, CAT, and GPX enzymes, which are pointers for OSI. However, AST indicated a P-value less than 0.01, which indicated a significant decrease in the serum concentrations of OSI and LDI with a 25mg/kg and 50mg/kg dose. Antioxidant therapy groups were also significantly enhanced by the extract and MDA concentration showed significantly decreasing values with increased SOD, CT and GPX. These findings suggest that the hydroalcoholic extract of Astragalus ecbatanus possesses potential hepatoprotective activity against repeated acetaminophen-induced liver injury in rats. Further mechanistic and clinical studies are required to confirm these findings.
